Testing will vary depending on the vehicle. Most electronic throttle bodies rest with the blade open enough for smoke to pass through, others will have to be manually opened either by scan tool or using the pedal. Opening the blade will certainly allow the smoke to fill the intake quicker.
If you suspect the throttle blade is causing a problem. I would suggest unplugging a vacuum line from the intake and check to make sure it is getting smoke through the line. If a good volume of smoke flows from the open vacuum port then you do not need to worry about the throttle blade.
